NSW Police will allege the incident was domestic violence-related, and the pair were known to each other.
Mid North Coast police inspector Vince Nicholls said forensic specialists were examining the Nambucca Heads property.
Police have not determined the cause or time of death of the 62-year-old man, he said.
“We are in contact with the family. Of course, they are distressed and upset at this time,” Nicholls said.
“Police do ask that anyone with information about the circumstances of this death please contact Crime Stoppers or Mid North Police or Coffs-Clarence Police.”
Gary Smith lives a few houses away from Nambucca Heads home.
He said he would not have expected an alleged murder in the area.
“He (the victim) just looked like a quiet, decent guy … looked after his yard and everything,” Smith said.
The 35-year-old woman been refused bail overnight and will appear at Port Macquarie Local Court on Saturday.
